About: Reactions of biological macromolecules span a wide time range, from femtosecond lifetimes of chemical intermediates to seconds or even longer for the overall reaction. The dynamic structures of the short-lived intermediates are extremely difficult to examine, although they are of critical relevance to understand the molecular mechanism at atomic level. To literally see how biomolecules work is therefore an objective for the biology and life science community in general. The strength of the European XFEL is its unique brilliance coupled with a high repetition rate bunch structure. This combination enables the investigation of biochemical reactions with high temporal and spatial resolution, finally resulting in so called “molecular movies”. However, a prerequisite for a successful outcome of these kinds of experiments is a diligent sample preparation and characterization. The XBI biolab of EuXFEL provides extensive onsite infrastructure and support for users, dedicated for this purpose. In this talk, an overview about various experimental possibilities for time-resolved experiments with biomolecules will be given. Particular emphasize will be put on the potential pitfalls and connected sample requirements. Relevant sample preparation and characterization strategies for selected examples, that have been investigated in the XBI so far, will be presented.
